추리 소설 역사상 가장 매력적인 인물 '셜록 홈스'를 창조해 전 세계 독자를 열광시킨 영국의 소설가이다. 1859년 5월 22일 영국 스코틀랜드 에든버러에서 태어났다. 아버지 찰스 얼터먼트 도일은 아일랜드계 잉글랜드인이었고, 어머니 메리 폴리는 아일랜드인이었다. 에든버러 대학에서 의학을 전공한 후 선박에서의 서부 아프리카 해안을 항해하는 등 의사 경험을 거쳐 포츠머스에서 개업하나 환자가 없어 소설을 쓰기 시작했다. 이 시기의 경험은 그의 소설에 폭넓은 소재와 주제를 제공했다.
그는 「사사싸 계곡의 미스터리」를 발표하면서 본격적으로 소설 쓰기를 시작했으며, 그러던 중 1887년에 셜록 홈즈가 등장하는 첫 작품 『주홍색 연구』를 발표했고, 1890년 두 번째 장편 『네 사람의 서명』을 발표하면서 점차 인기가 높아졌다. 1891년 런던에서 다시 개업하지만 역시 성공하지 못했기에 작품에 전념하기로 결심하고 1892년에 『셜록 홈즈의 모험』과 『셜록 홈즈의 회상』(1894) 등 홈즈 시리즈 단편을 차례차례로 발표하여 추리소설의 장르를 확립했다. '셜록 홈즈' 시리즈만으로 두 편의 장편과 네 권의 단편집을 발표하였다. 냉정하고 날카로운 홈즈와 온후한 왓슨이 여러 사건에 도전하는 이 시리즈는 60여 편에 이른다.
셜록 홈스 이야기는 처음 발표되자마자 세상에 돌풍을 일으켰고 세계 각국에 소개되었다. 독자들은 괴팍한 성격과 탁원한 재능으로 카리스마를 풍기는 홈스의 모습에 매료되었다. 그 결과 홈스는 명탐정의 대명사가 되었고, 심지어 많은 독자가 그를 실제 인물이라고 믿기까지 했다. 『용감한 제랄의 모험담』, 『잃어버린 세계』 등의 과학소설도 썼다. 1902년, 보어 전쟁에서 의사로 활약, 영국의 참전을 정당화하는 등의 업적으로 기사 작위에 서임되었다. 제1차 세계대전에서 아들을 잃은 후 심령현상에 관심을 보였다.
홈즈 시리즈가 준 영향은 탐정소설에만 국한되지 않았다. 셜로키언이라 불리는 팬이 전 세계에 존재한다. 40년의 세월 동안 꾸준히 홈스 시리즈를 발표하며 미스터리의 보급에 기여했다. 이후 애거서 크리스티, 도러시 세이어스, 앤서니 버클리, S.S.밴 다인 등의 작가들이 등장하는 데 발판이 되어 주었다. 이후에도 아서 코난 도일은 꾸준히 미스터리 장르 작품 활동에 매진하였으나 1930년 7월 7일, 심장마비로 사망하였다.
